Harlem Hospital School of Nursing was a training school for African-American women, which was established at Harlem Hospital in Harlem, New York City in 1923. It was founded due to the lack of nursing schools in New York that accepted African American women. Until 1923, the Lincoln Hospital School for Nurses in The Bronx was the only school that allowed the enrollment of Black women. When Mayor Hylan sought reelection in 1921, the NAACP and other community organizations lobbied the mayor to improve healthcare access. Around the same time, Lurline Vassall of Brooklyn, was denied entry to the Bellevue Hospital School of Nursing because of her race. Lurline's father William Vassall launched a campaign to open a school for black nurses. In response, Hylan's administration supported the creation of the Harlem Hospital School of Nursing.The school opened on January 3, 1923, with a class of twenty black women. It was a two and a half year program.The Harlem Hospital School of Nursing closed in 1977.

The Savoy Ballroom was a large ballroom for music and public dancing located at 596 Lenox Avenue, between 140th and 141st Streets in the Harlem neighborhood of Manhattan, New York City. Lenox Avenue was the main thoroughfare through upper Harlem. Poet Langston Hughes calls it the "Heartbeat of Harlem" in Juke Box Love Song, and he set his work "Lenox Avenue: Midnight" on the legendary street. The Savoy was one of many Harlem hot spots along Lenox, but it was the one to be called the "World's Finest Ballroom". It was in operation from March 12, 1926, to July 10, 1958, and as Barbara Englebrecht writes in her article "Swinging at the Savoy", it was "a building, a geographic place, a ballroom, and the 'soul' of a neighborhood". It was opened and owned by white entrepreneur Jay Faggen and Jewish businessman Moe Gale. It was managed by African-American businessman and civic leader Charles Buchanan. Buchanan, who was born in the British West Indies, sought to run a "luxury ballroom to accommodate the many thousands who wished to dance in an atmosphere of tasteful refinement, rather than in the small stuffy halls and the foul smelling, smoke laden cellar nightclubs ..."

Harlem Hospital Center, branded as NYC Health + Hospitals/Harlem, is a 272-bed, public teaching hospital affiliated with Columbia University. It is located at 506 Lenox Avenue in Harlem, Manhattan, New York City and was founded in 1887.The hospital was established to provide healthcare to the citizens of the neighborhood. Initially, the hospital served as a holding area for patients to be transferred to Randalls and Wards Islands and Bellevue Hospital. With the wave of the African Americans who moved to New York after World War I, the hospital soon outgrew its initial building. After acquiring land, a new building opened on April 13, 1907. The hospital developed a teaching program that is affiliated with Columbia University, and has continued to serve the Harlem neighborhood since its inception.

The Dark Tower was a townhouse on 108 West 136th Street in Harlem.

The Lincoln Theatre is a theater located on 135th Street near Lenox Avenue in Harlem, New York City. It opened in 1915 and was the first theater in a then predominantly white neighborhood in Harlem to cater to black audiences. The theater reached its peak of fame in the 1920s, when entertainers such as Bessie Smith, Florence Mills, and Fats Waller headlined. (Fats Waller had been hired as the organ player of the theater when he was fifteen years old.) The Lincoln Theatre was the only place in New York where Ma Rainey performed.

The Schomburg Center for Research in Black Culture is a research library of the New York Public Library (NYPL) and an archive repository for information on people of African descent worldwide. Located at 515 Malcolm X Boulevard (Lenox Avenue) between West 135th and 136th Streets in the Harlem neighborhood of Manhattan, New York City, it has, almost from its inception, been an integral part of the Harlem community. It is named for Afro-Puerto Rican scholar Arturo Alfonso Schomburg. The resources of the center are broken up into five divisions, the Art and Artifacts Division, the Jean Blackwell Hutson General Research and Reference Division, the Manuscripts, Archives and Rare Books Division, the Moving Image and Recorded Sound Division, and the Photographs and Prints Division. In addition to research services, the center hosts readings, discussions, art exhibitions, and theatrical events. It is open to the general public.

The 135th Street station is a station on the IRT Lenox Avenue Line of the New York City Subway. Located at the intersection of 135th Street and Lenox Avenue in Harlem, it is served by the 2 and 3 trains at all times. The 135th Street station was constructed for the Interborough Rapid Transit Company (IRT) as part of the city's first subway line, which was approved in 1900. Construction on the tunnel segment that includes the 135th Street station started on October 2 of the same year. The station opened on November 23, 1904. The station platforms were lengthened in 1910. The 135th Street station contains two side platforms and three tracks. The station was built with tile and mosaic decorations. The platforms contain exits to Lenox Avenue's intersection with 135th Street and are not connected to each other within fare control. The platforms contain elevators from the street, which make the station compliant with the Americans with Disabilities Act of 1990.

The Abyssinian Baptist Church is a Baptist megachurch located at 132 West 138th Street between Adam Clayton Powell Jr. Boulevard and Lenox Avenue in the Harlem neighborhood of Manhattan, New York City. It is affiliated with the National Baptist Convention, USA and American Baptist Churches USA. Its senior pastor is Johnnie D. Brooker, Jr.. The Abyssinian Baptist Church congregation traces its history to 1809, when seamen from the Ethiopian Empire (then known as Abyssinia) helped lead a protest against racially segregated church seating. Thomas Paul was an early minister. During the 20th century, prominent ministers of the church included Adam Clayton Powell Sr., Adam Clayton Powell Jr., and Samuel DeWitt Proctor. The church has served as an influential place for African-American spirituality, politics and community. The church worshiped in several places before building the present church structure Its present building was built in 1922–23 and was designed by Charles W. Bolton & Son in Gothic Revival and Tudor Revival styles – it has also been described as "Collegiate Gothic". It features stained glass windows and marble furnishings. The church and its associated community house were designated a New York City Landmark on July 13, 1993.

The Mother African Methodist Episcopal Zion Church, also known as "Mother Zion", located at 140–148 West 137th Street between Adam Clayton Powell Jr. Boulevard and Lenox Avenue in the Harlem neighborhood of Manhattan, New York City, is the oldest African-American church in New York City, and the "mother church" of the African Methodist Episcopal Zion conference.The church was designated a New York City Landmark on July 13, 1993.

The Harlem YMCA is located at 180 West 135th Street between Lenox Avenue and Adam Clayton Powell Jr. Boulevard in the Harlem neighborhood of Manhattan, New York City. Built in 1931-32, the red-brown brick building with neo-Georgian details was designed by the Architectural Bureau of the National Council of the YMCA, with James C. Mackenzie Jr. as the architect in charge. It replaced the building from 1919 across the street. Inside the building is a mural by Aaron Douglas titled "Evolution of Negro Dance." The building was declared a National Historic Landmark in 1976, and was designated a New York City Landmark in 1998.

The Cotton Club was a New York City nightclub from 1923 to 1940. It was located on 142nd Street and Lenox Avenue (1923–1936), then briefly in the midtown Theater District (1936–1940). The club operated during the United States' era of Prohibition and Jim Crow era racial segregation. Black people initially could not patronize the Cotton Club, but the venue featured many of the most popular black entertainers of the era, including musicians Fletcher Henderson, Duke Ellington, Jimmie Lunceford, Chick Webb, Louis Armstrong, Count Basie, Fats Waller, Willie Bryant; vocalists Adelaide Hall, Ethel Waters, Cab Calloway, Bessie Smith, Aida Ward, Avon Long, the Dandridge Sisters, the Will Vodery Choir, The Mills Brothers, Nina Mae McKinney, Billie Holiday, Midge Williams, Lena Horne, and dancers such as Katherine Dunham, Bill Robinson, The Nicholas Brothers, Charles 'Honi' Coles, Leonard Reed, Stepin Fetchit, the Berry Brothers, The Four Step Brothers, Jeni Le Gon and Earl Snakehips Tucker. At its prime, the Cotton Club served as a hip meeting spot, with regular "Celebrity Nights" on Sundays featuring guests such as Jimmy Durante, George Gershwin, Sophie Tucker, Paul Robeson, Al Jolson, Mae West, Richard Rodgers, Irving Berlin, Eddie Cantor, Fanny Brice, Langston Hughes, Judy Garland, Moss Hart, and Jimmy Walker, among others.

The Renaissance Ballroom & Casino was an entertainment complex at 2341–2349 Adam Clayton Powell Jr. Boulevard (Seventh Avenue) in the Harlem neighborhood of Manhattan in New York City. When opened in 1921, it included a casino, ballroom, 900-seat theater, six retail stores, and a basketball arena. It spanned the entire eastern frontage of Adam Clayton Powell Jr. Boulevard between 137th and 138th Streets.
